With the return of the Primarch Roboute Guilliman, the Imperium of Man initiates the galaxy-spanning Indomitus Crusade, a relentless ten-year endeavor aimed at reclaiming lost territories from the grasp of its adversaries and salvaging it from the precipice of collapse. Yet, just as victory seems within reach, a formidable new threat emerges from the depths of the western galaxy—the Tyranids of Hive Fleet Leviathan, swarming in unprecedented numbers, reigniting the flames of conflict in what becomes known as the 4th Tyrannic War.

Warhammer 40,000: Space Marine 2 pits players against two primary adversaries: the Tyranids, believed to be a splinter of Hive Fleet Leviathan, and the forces of Chaos. The Tyranids, an extragalactic insectoid species driven by an insatiable hunger for biomatter, pose an existential threat to all life. Meanwhile, the forces of Chaos, followers of the malevolent Gods of Chaos, make their malevolent return, with the Thousand Sons, disciples of Tzeentch, the Chaos God of Sorcery, Manipulation, and Mutation, confirmed to play a pivotal role.

Following the events of the preceding game, Captain Titus (now voiced by Clive Standen) finds himself embroiled in an investigation by the Inquisition, accused of heresy. Although eventually exonerated and permitted to rejoin the Ultramarines Chapter, lingering suspicion among his comrades leads to his demotion to the rank of Lieutenant. Subsequently, Titus undergoes the Rubicon Primaris, a transformative process elevating him from a Firstborn Marine to a Primaris Space Marine, endowed with heightened strength, speed, and stature.

The narrative thrust of the main campaign follows Lieutenant Titus as he leads the charge against the encroaching forces of Hive Fleet Leviathan, which have besieged the Imperial worlds of Avarax and Kadaku. Assisted by his new squadmates, Brother-Sergeant Gadriel and Brother Chairon, Titus confronts the relentless onslaught of the Tyranids, battling to secure the survival of the Imperium against overwhelming odds.
